چکیده

The a7 subtype of the nicotinic acetylcholine receptor (nAChR) is the target of studies aimed at identifying features that will lead to the development of selective therapeutics. Five arylidine anabaseines, three with pyridine rings and two with the pyrrole rings, were synthesized in 35–65% yield via aldol condensation. The compounds are homologs of benzylidine anabaseine and were chosen for synthesis because they provide either a hydrogen bond acceptor (pyridines) or hydrogen bond donor (pyrroles) that may interact with the receptor within the benzylidine selectivity motif. Initial analysis of the new compounds at 100 lM concentration reveal that the two pyrrole anabaseines are good partial agonists of the a7 nAChR, having 40% of the efficacy of ACh, efficacy comparable to 4OH-GTS-21, and dramatically enhanced efficacy relative to the 2and 4-pyridinyl compounds. The pyrrole compounds were confirmed to be a7 selective, displaying preference for this receptor over muscle and heteromeric neuronal receptor

Ivermectin: A Positive Allosteric Effector of the a7 Neuronal Nicotinic Acetylcholine Receptor

We report that preapplication of ivermectin, in the micromolar range, strongly enhances the subsequent acetylcholine-evoked current of the neuronal chick or human a7 nicotinic acetylcholine receptors reconstituted in Xenopus laevis oocytes and K-28 cells. This potentiation does not result from nonspecific Cl currents. The concomitant increase in apparent affinity and cooperativity of the dose-r...

The Novel a7b2-Nicotinic Acetylcholine Receptor Subtype Is Expressed in Mouse and Human Basal Forebrain: Biochemical and Pharmacological Characterization s

We examined a7b2-nicotinic acetylcholine receptor (a7b2nAChR) expression in mammalian brain and compared pharmacological profiles of homomeric a7-nAChRs and a7b2-nAChRs. a-Bungarotoxin affinity purification or immunoprecipitation with anti-a7 subunit antibodies (Abs) was used to isolate nAChRs containing a7 subunits from mouse or human brain samples. a7b2-nAChRs were detected in forebrain, but ...

Atypical nicotinic agonist bound conformations conferring subtype selectivity.

The nicotinic acetylcholine (ACh) receptor (nAChR) plays a crucial role in excitatory neurotransmission and is an important target for drugs and insecticides. Diverse nAChR subtypes with various subunit combinations confer differential selectivity for nicotinic drugs. We investigated the subtype selectivity of nAChR agonists by comparing two ACh-binding proteins (AChBPs) as structural surrogate...

Nicotine Stimulates PPARB/D Expression in Human Lung Carcinoma Cells through Activation of PI3K/mTOR and Suppression of AP-2A

We previously showed that nicotine stimulates non–small cell lung carcinoma (NSCLC) cell proliferation through nicotinic acetylcholine receptor (nAChR)–mediated signals. Activation of peroxisome proliferator–activated receptor B/D (PPARB/D) has also been shown to induce NSCLC cell growth. Here, we explore the potential link between nicotine and PPARB/D and report that nicotine increases the exp...
